Based on market research, Iacocca saw that younger buyers were beginning to dominate the market. The design and appearance of a new model was critical. Cars coming off the Ford assembly lines had to be more than reliable, efficient and functional. They had to look good, what Iacocca described as “a car you could drive to the country club on Friday night, to the drag strip on Saturday, and to church on Sunday.”The result was the Mustang.Months in advance of its debut, an aggressive promotional campaign was launched. Editors of college newspapers were loaned Mustangs to drive several weeks before the official introduction date. A massive recall was ordered.In addition, rising fuel costs during the Arab oil embargo and increasing competition from the Japanese and other foreign car industries that specialized in sporty and fuel-efficient vehicles were threatening Detroit.Iacocca’s eight years as president were made difficult by his tense relationship with Henry Ford II.As Iacocca told it in his autobiography, Ford repeatedly reduced the president’s authority and prestige through a series of executive reorganizations, directed the arbitrary and capricious firing of other executives, and had a close friend of Iacocca’s investigated for Mafia connections, which were never found.“If a guy is over 25 percent a jerk, he’s in trouble,” Iacocca later said. “And Henry was 95 percent.”He said Ford never gave a specific reason for his dismissal. “We had just completed the two best years in our history,” he wrote in his autobiography. But he quoted Ford as telling him: “I think you should leave. It’s personal. . . . It’s just one of those things . . . sometimes you just don’t like somebody.”The Ford-Iacocca showdown was widely perceived as a dust-up between the upstart son of Italian immigrants and the scion of the automotive aristocracy. That Iacocca would cast his lot with Chrysler, a Ford competitor, after being fired was seen as a case of “Don’t get mad. Get even,” which only enhanced the Iacocca mystique.After Iacocca, then 54, accepted an offer to run Chrysler, found a company badly mismanaged – “Nobody knew who was on first,” he said – and hemorrhaging cash.John Riccardo, Iacocca’s predecessor as the Chrysler chairman, had already been traveling to Washington seeking help from Congress in the form of tax credits and the easing of federal regulations, but Riccardo had won little support.Only as a last resort, Iacocca said, did the company ask for federal loan guarantees. The exposure he received through these appearances and Chrysler commercials propelled Iacocca into a national celebrity.The architect of this effort was Leo-Arthur Kelmenson, chief executive of what then was Kenyon & Eckhardt, the advertising agency that had at great risk quit its long-standing client, Ford, to work for Chrysler at Iacocca’s behest.Kelmenson devised an ad campaign that asked bluntly, “Would America be better off without Chrysler?” The answer clearly was no, the ad suggested.Also from Kelmenson came the idea to feature Iacocca in the television commercials for Chrysler cars. It was rare, if not unprecedented, for such advertisements to feature chief executives in person, but Kelmenson argued that television footage of Iacocca in the flesh would give the commercials a credibility and heft they could gain in no other way.There was an element of patriotism in the Iacocca message. By the 1980s, the American automobile industry was no longer the globally dominating colossus it had been. Foreign cars had established deep inroads. Iacocca liked to mention that the cars he was selling were made in America, which conveyed a less-than-subliminal thought that to buy a Chrysler product was to strike a blow on behalf of the U.S.A. against foreign intrusion.In addition to aggressive cost-cutting measures, Iacocca boosted Chrysler finances with strong sales of the Dodge Caravan and Plymouth Voyager minivans. On those sales and the acquisition of American Motors in 1987 – the maker of the Jeep brand – lay the foundation on which Iacocca rebuilt Chrysler.The firm had a long history teetering between sizable profits and losses, and Iacocca could never entirely keep it in the black. He retired in 1992 but remained a consultant at $500,000 a year.In 1995, he backed billionaire investor Kirk Kerkorian’s unsuccessful $23 billion hostile takeover bid for Chrysler. This led to an attempt by the car company to block Iacocca from exercising $42 million in stock options, claiming he gave away confidential information. He sued, and the claim was settled out of court for $21 million.Iacocca’s first wife died in 1983. Iacocca’s later marriages, to advertising executive Peggy Johnson and restaurateur Darrien Earle, ended in divorce. In addition to his daughter, of Laguna Beach, California, survivors include another daughter from his first marriage, Kathryn “Kate” Iacocca Hentz of Cohasset, Massachusetts; a sister; and eight grandchildren.In retirement, Iacocca remained outspoken about the business he knew best.He was dismissive of Chrysler’s merger in 1998 with the German automaker Daimler-Benz, a union that fell apart less than a decade later amid a clash of corporate cultures. Chrysler was sold to the private equity firm Cerberus Capital Management, which in 2009 allowed Italian carmaker Fiat a large minority stake in Chrysler.Even in later years, Iacocca maintained an aura of success. In recent years, he was often invited back to Chrysler give pep rallies as the company continued to cut jobs and suffer amid rising gas prices, economic turmoil and plummeting sales.He also returned to serve as pitchman, helping the company appeal to different demographics by appearing in commercials with “Seinfeld” actor Jason Alexander and the rapper Snoop Dogg.In one TV spot, Iacocca’s famous pitch line – “If you can find a better car, buy it!” – gets a hip-hop translation by Snoop Dog: “If the ride is more fly, then you must buy.” When Iacocca tells viewers the car is a good deal, Snoop Dogg replies, “Fo shizzle, I-ka-zizzle.”“He’s just a good kid,” Iacocca told USA Today about his co-star. “I didn’t understand half the things he was telling me, but it was fun.” 8 Comments Almost immediately, Iacocca rebounded as chairman of Chrysler. You may recall that Brian Jenkins, the DIY Guy over at i1Tesla, was the first to bring us a simple and inexpensive modification to fully open the Model 3 frunk hands-free. The latest creation from Jenkins’ lab is a Tesla Model 3 modification that allows objects as long as 10 feet to be hauled inside the modestly-dimensioned Model 3. What is commonly known as a “Ski Pass-Thru” is a small opening between the trunk and the rear passenger seating area permitting long but narrow cargo (like snow skis or 2×4 lumber) to fit completely inside the vehicle. “I wasn’t sure it was even possible, but it was an interesting feature request from an i1Tesla member that I thought should be explored”, Jenkins said.Jenkins was the first to perform this customization on his own Tesla Model 3. He even put the entire project on video for the world to see how it’s done. While he admits this DIY project isn’t for most people due to the complexity (and the fact that you have to cut through the steel seat back plate), he wanted to demonstrate that it was indeed possible. The i1Tesla video shows step-by-step the removal of the rear seat, the customization of the backrest, and the finished installation. The pass-thru feature remains hidden behind the rear seat folding armrest when not in use keeping the interior appearance factory stock. Simply folding down the armrest reveals the pass-thru directly into the trunk space. If you need to pick up some lumber, haul snow skis, or some other long (but narrow) item you no longer need to take the pickup truck. Just jump into your Model 3 and get the job done quickly.Brian has plans to do more of these unique DIY projects on his channel to demonstrate the possibilities for the Tesla Model 3, and to help owners make practical improvements that save time and money. The C-HR and IZOA will be the first BEVs to launch in China under the Toyota brand. The most notable was its compact crossover, the C-HR EV. It’s the first completely electrified vehicle from the Japanese brand to come to China. Curiously, the other main EV is also the C-HR, but wears a badge proclaiming itself to be the Izoa EV. It, according to Autocar,  will be built by the state-owned outfit FAW. Toyota says the twins should be offered to consumers in 2020.More about Toyota in China: The third vehicle drives solidly on the concept-vehicle side of the road. The Rhombus (image below, as well as a brief video) was designed by TMEC (Toyota Motor Engineering & Manufacturing (China)). Besides the soft quadrilateral shapes on each side of its protruding snout, the geometric shape repeats inside. The seats are laid out in a rhombic fashion, which gives all four seating positions tons of leg space for stretching out. This helps the car keep a narrow profile, but unfortunately also removes any meaningful amount of cargo space.The appearance of the all-electric C-HR sparks the inevitable questions, “Will it come to other markets?” While we have no definitive answer, the automaker reminds us that it plans to introduce more than 10 battery-electric vehicles before 2025. While we would hope it makes its way outside of China, the automaker has yet to release technical specifications for the vehicle, so it’s difficult to judge how well suited it might be for Western markets. first_imgBecause Senator Chuck Grassley is a Republican and Hillary Clinton is a Democrat, it would be easy to dismiss the underlying issues as merely political gamesmanship.However, if that is your reaction to this recent letter from Senator Grassley to U.S. Attorney General Loretta Lynch (and others) it would be unfortunate because the underlying issues go to the glaring double standard that has frequently been highlighted on FCPA Professor.The double standard being how business interactions with “foreign officials” seem to be subject to different standards than business interactions with U.S. officials.Senator Grassley’s letter concerns allegations first raised in this New York Times article (see also here) concerning the “Clinton Foundation’s tie to a number of investors involved in a business transactions that resulted in the acquisition of Uranium One, owner of U.S. based Uranium assets, by a subsidiary of Rosatom, a Russian government owned company.As stated in Senator Grassley’s letter:“[D]uring critical stages of the acquisition approval, interested parties made large donations – some in the millions of dollars – to the Clinton Foundation while Ms. Hillary Clinton held the position of Secretary of State. When millions of dollars flow to decision makers who have substantial discretion to provide support for or against approval of controversial transactions, public confidence in the integrity of the process requires a commitment to transparency and responsiveness to oversight inquiries.”The Uranium One deal is merely one of several criticisms or concerns of financial ties between the Clinton Foundation and various business interests.  See here, here, here, here, here, here, here, here, here and here. first_imgJun 21 2018A team of researchers led by Dr Pierre Close, WELBIO researcher at the ULiège GIGA Institute and Dr Francesca Rapino has uncovered a new therapeutic opportunity in the treatment of malignant melanoma that acquired resistance to targeted therapies. In collaboration with researchers from VIB, they have revealed that malignant melanoma can reprogram their protein synthesis machinery and become addicted to a new family of enzymes that modify transfer RNAs during acquired resistance. Strikingly, the inhibition of these molecules synergies with targeted therapies to produce a strong anti-tumoral effect. These new findings, published in Nature, will be key in the development of improved diagnostic tools and melanoma treatment.Resistance to therapy is the principal limitation of current treatment of aggressive cancers such as malignant melanoma. Insurgence of resistance relates to the capability of tumor cells to circumvent the stress induced by the treatment. In order to survive, cancer cells develop a series of adaptation mechanisms through rewiring fundamental processes. Among those, reprogramming of mRNA translation favors the expression of proteins essential for tumor development. The lab of Dr. Close has been studying the contribution of wobble tRNA modification in cancer development through regulation of selective mRNA translation for a few years now, uncovering their central role in tumor initiation and metastatic potential.Modification of certain tRNA molecules at the wobble position regulates selective mRNA translation and impact on protein expression. The GIGA-ULiège team discovered that melanoma that carry the BRAF(V600E) mutation – found in more than 50% of the melanoma patients – are addicted to enzymes modifying wobble uridine tRNAs (U34-tRNA).Francesca Rapino, post-doctoral researcher in Dr Close’s lab: “Our research showed that wobble uridine tRNA modification enzymes are upregulated in melanoma clinical samples and very lowly expressed in melanocytes, the normal melanin-producing cells. Strikingly, inhibition of this family of enzymes led to a very strong and specific cell death in BRAF(V600E) melanoma, but had no effect on melanocytes. Therefore, the inhibition of these enzymes prevents glucose metabolism in melanoma cells, and limits their energy income. As a consequence, the growth and survival of melanoma cells is strongly reduced after inhibition of U34-tRNA enzymes. Strikingly, we discovered that acquired resistance to targeted therapy, which strongly limits the clinical benefit of the treatment of malignant melanoma, is prevented by the inhibition of U34-tRNA enzymes. In other words, the inhibition of these enzymes synergizes with targeted therapies to block malignant melanoma growth.”This work revealed the clinical potential of U34-tRNA enzymes inhibition for the treatment of human malignant melanoma, a disease that remains extremely difficult to treat. The findings may provide important information for identifying which patients potentially fare better when treated with an alternate therapy.In a newly published study in Journal of Clinical Investigation, researchers found that abiraterone, a common prostate cancer drug, yields high-levels of a testosterone-like byproduct in men with advanced disease who have a specific genetic variant.The study’s lead researcher, Nima Sharifi, M.D., Cleveland Clinic Lerner Research Institute Department of Cancer Biology, previously discovered that men with aggressive prostate cancer who have a specific variant in the HSD3B1 gene have poorer outcomes than patients without the variant. HSD3B1 encodes an enzyme that allows cancer cells to use adrenal androgens for fuel. This enzyme is overactive in patients with the variant HSD3B1(1245C).In the new study, Dr. Sharifi and his team, including first author Mohammad Alyamani, Ph.D., found that men with the genetic anomaly metabolize abiraterone differently than men without the variant. “We are hopeful these finding will lead to us being able to better tailor prostate cancer treatments based on a patient’s specific genetic make-up,” said Dr. Sharifi. “More studies are needed, but we have strong evidence that HSD3B1 status affects abiraterone metabolism and probably its effectiveness. If confirmed, we hope to identify an effective alternative drug that might be more effective in men with this genetic anomaly.”Traditional therapy for advanced prostate cancer – called androgen deprivation therapy (ADT) – blocks cells’ supply of androgens (male hormones), which they use to grow and spread. While ADT is successful early on, cancer cells grow resistant, allowing the disease to progress to a lethal phase called castration-resistant prostate cancer (CRPC). In CRPC, cancer cells utilize an alternative source of androgens produced in the adrenal glands. The metabolite tricks androgen receptors into turning on dangerous pro-cancer pathways. Remarkably, this byproduct of abiraterone – which is designed to block androgens – may behave like an androgen and cause prostate cancer cells to grow. Investigating abiraterone’s impact on clinical outcomes in CRPC patients will be an important next step.”This study adds to our understanding of the deleterious effect of inherited variants of the HSD3B1 gene and holds promise for precision medicine approaches in the management of men with advanced prostate cancer,” said Eric Klein, M.D., chair of Cleveland Clinic Glickman Urological and Kidney Institute.The study was supported in part by grants from the National Cancer Institute and Prostate Cancer Foundation.”This study helps to define a novel resistance pathway for abiraterone, a commonly used medication for patients with advanced prostate cancer. first_imgJun 27 2018Heart disease is the number one killer in the United States, and high triglyceride levels in the blood are cited as just one of several risk factors. Millions of lipid panels, blood tests that look at cholesterol levels as well as triglycerides, are performed in clinics each year.Two new Morgridge Institute for Research studies suggest the current tests, which measure the abundance of lipid classes, are insufficient. Rather, lipids identified and studied at the individual species level–instead of grouped in classes–may be better signatures of metabolic health.The results were published online June 13, 2018, in Cell Systems as open access papers, one focusing on plasma lipid species and the second on liver lipid species.Lipids, or fats, are incredibly important to human health, yet one of the hardest biomolecules to study. Harnessing advances in mass spectrometry technology, Morgridge scientists measured almost 150 lipid species in the blood and liver of mice and identified some that can act as signatures of healthy or unhealthy metabolic states.For patients getting a lipid panel–something the American Heart Association suggests for everyone over the age of 20–results will include a reference to triglyceride levels. These tests look at triglycerides in bulk, as a group, and measure how much is in the blood.But Molly McDevitt, a graduate student in Dave Pagliarini’s Lab at Morgridge and co-first author on the papers, says looking at individual species of triglycerides provides a much more accurate picture.”We don’t even know how many different triglycerides there are–hundreds, thousands,” says McDevitt. “We found that some triglycerides correlate positively with a fatty liver, while others correlate negatively with a fatty liver. Often, in order to identify a fatty liver, an invasive liver biopsy is required. Taking a blood sample would be a much simpler way to diagnose it.The studies are still in the early stages, but the results look promising.”As we move from measuring a bulk lipid class in serum to specific lipids, we’re finding that some do indeed predict what’s happening in the liver,” says Dave Pagliarini, director of the Morgridge Metabolism Theme. “That gives us confidence that we might be able to discover biomarkers in plasma that report on what’s happening in organ metabolism.”Using multi-omics approaches to further discoveryBy combining lipids data with other biomedical datasets, an approach known as multi-omics, the scientists were able to identify and prioritize sets of genes they know are causing the changes in lipid levels.”We might find a location on the DNA that contains 400 genes, and it would be really hard to follow up on all of those,” McDevitt says. “By using the different layers of the multi-omic approach, like the phenotypes and the gene expression levels, it helps us narrow down that list to something much easier to handle.”Future work will investigate regions of the genome the team found linked to the various lipids. first_imgThe gargantuan mastodons, mammoths, and giant ground sloths that once roamed North America seem much too big for most prehistoric predators to contend with. But a new study suggests that the lions and saber-toothed cats that once roamed North America did indeed attack these beasts and may have significantly reduced their populations. If so, these carnivores may have had a much more dramatic impact on ancient ecosystems than previously believed.“This is a landmark paper because it really puts together an unusually broad range of lines of evidence on this issue,” says John Damuth, paleobiologist at the University of California (UC), Santa Barbara, who was not involved in the study.The scientific thinking that large herbivores are immune to predators goes back to Charles Darwin. In his 1859 Origin of Species, the famed naturalist wrote that “… with the elephants and rhinoceroses, none are destroyed by beasts of prey. Even the tiger in India most rarely dares to attack a young elephant protected by its dam.” But Blaire Van Valkenburgh wasn’t so sure. “I just said, no, I don’t believe that,” says the UC Los Angeles paleoecologist and lead author of the new study. Required fields are indicated by an asterisk (*)center_img Email To find out what ancient predators were capable of, Van Valkenburgh and her colleagues first had to figure out just how big the large herbivores and carnivores were that lived during the Pleistocene epoch, 12,000 to 2.5 million years ago. The researchers used thousands of measurements of fossil bones and compared them to the bones of modern elephants to infer the range of body masses of both adults and juveniles of the different species of extinct megaherbivores, including mammoths and mastodons. They used a similar method to infer the size ranges of  extinct carnivore species, comparing saber-toothed cats to modern tigers, for example.The analyses indicated that the juvenile megaherbivores weighed from 200 to 2000 kg. Adults weighed several tons; estimates for some adult males exceed 10,000 kg. The prehistoric carnivores ranged from 150 kg to more than 400 kg.So could ancient carnivores have actually taken down these herbivores? To answer this question, the researchers compiled information from thousands of records of prey killed by big modern carnivores such as lions and tigers hunting solo and in groups—a task that took several years. Conservation ecologist and co-author Matt Hayward of Bangor University in the U.K. used these data to develop a mathematical model to predict typical and maximum prey size for any given carnivore.Finally, the researchers needed to determine how protected young mega-herbivores were. “There’s a sweet spot in there, from the predator’s standpoint, where the juveniles can wander away from mom frequently enough and far enough to be accessible, and where they are of a size small enough that they can actually be tackled,” explains co-author Louise Roth, a biologist at Duke University in Durham, North Carolina.For this, the researchers relied on earlier studies of the chemical signature in the enamel layers that are added annually at the base of herbivore tusks as they grow out from the skull. The chemistry of the tusks changes when the animal’s diet switches from mother’s milk to vegetation. The researchers assumed that baby herbivores stayed close to mom while nursing, but wandered farther away once they began to forage for themselves. The team found that there was indeed a window when juvenile mammoths and other herbivores were weaned but were still small enough to be hunted by lions and other predators. “A solitary saber-toothed cat could have taken young mammoths aged 2 to 4—sometimes up to age 9—and a pride would have been able to take adult females,” says Roth.  All told, lions, wolves, and saber-toothed cats could have killed about 17% of young mastodons and other mega-herbivores, the team reports online today in the Proceedings of the National Academy of Science [url tk]. Although the researchers do not have direct evidence of predation on the bones of the extinct herbivores, for studies of prehistoric ecosystems, “getting smoking gun evidence is always difficult,” notes Kathleen Lyons, paleobiologist at the Smithsonian Museum of Natural History who was not involved in the study. But the evidence that Van Valkenburgh and her coauthors brought together to make the case is convincing, she says. “It’s a really nice, solid, piece of work.”These predators didn’t just have dramatic impacts on prey populations—they could have had cascading effects on the entire ecosystem, Van Valkenburgh says. “Large carnivores provide carcasses that feed a whole array of smaller species like eagles, coyotes, and foxes,” she says. And keeping the megaherbivore populations in check meant that there was more vegetation for small mammals and birds. The predators might even have indirect effects on river ecosystems, she says, because the banks of the rivers were not being denuded by megaherbivores and eroding rapidly. “So aquatic species like beavers and otters did well and fish were abundant.”The work may have important implications for today’s conservation efforts, Damuth says. “It’s important to know that the current ecological communities differ in critical ways from those in which those herbivores evolved,” he says. Modern megaherbivores like elephants and rhinoceroses are threatened, and “if we want to manage them, we have to understand how they worked naturally, as opposed to how they might be working under current conditions.”last_img read more

first_imgA new microscope has given researchers their most detailed view yet of the earliest days of mouse development. For the first time, scientists can track each cell as it divides, from the fertilized egg all the way to the round ball of cells called a blastocyst, which implants in the mother’s uterus (see video, above). Mammalian embryos, including mice and humans, are very sensitive to the bright light used in standard high-resolution microscopes. A technique called light-sheet microscopy illuminates just a thin slice of the embryo at a time, minimizing the living specimen’s exposure. Researchers have now designed a light-sheet microscope that can image multiple embryos at once, keeping them at a stable temperature in their growth media. That allows the scientists to average out the differences between individual embryos and better recognize meaningful patterns in development. They report this week in Nature Methods that in mouse embryos, the first differentiation of cell types happens when the embryo grows from eight to 16 cells. When the cells in the eight-celled embryo divide, cells that end up on the inside (tagged red in this video) continue to develop into the body of the mouse, while those on the outside (tagged blue) go on to form the placenta. Not all species sleep alike, however, and scientists have long puzzled over which aspects are truly fundamental. Now, a new study on lizards suggests that sleep states once thought to occur only in mammals and birds have much older evolutionary origins.Scientists had long suspected that birds and mammals are the only vertebrates to experience rapid eye movement (REM), a sleep state in which the body is mostly immobile but the brain is in overdrive. During REM sleep, the brain generates high-frequency waves of electrical activity and the eyes flicker; in humans, REM is closely linked to dreaming. Punctuating REM are interludes of slow-wave sleep, a state in which brain activity ebbs and the waves become more synchronized. This slower state is widely thought to be important to memory formation and storage.But scientists who looked for signs of REM and slow-wave sleep in reptiles have had “confusing” results, says Gilles Laurent, a neuroscientist at the Max Planck Institute for Brain Research in Frankfurt, Germany. So when he and colleagues picked up on similar sleep patterns in Australian dragons (Pogona vitticeps) while recording their brain activity for a separate study, it came as a shock.center_img The team had planned to examine how the lizards—a common pet in Germany—use visual information to chase treats. They continuously recorded the lizards’ brain activity with electrodes over several weeks. At night, the sleeping reptiles’ brains produced rhythms that could be separated into two different patterns—one at very low frequency, about 4HZ, and another, higher frequency about 20HZ, the team reports today in Science. The two frequencies alternated every 40 seconds, reminding Laurent of the regular oscillations between high-frequency REM and slow-wave sleep found in mammals and birds. “The more we looked, the more it appeared as though we were looking at bona fide REM sleep,” he says.Using an infrared camera, the team found that the sleeping lizards’ eyelids twitched during the REM-like stage, just like other animals. They also found a tantalizingly familiar pattern within the slower phase of the lizards’ brain waves. During this slow phase, electrodes picked up sharp waves of voltage, followed by ripples of electricity that closely resembled patterns seen in humans and rodents. Some scientists believe these waves and ripples help convert new information into memories by replaying past events in fast-forward. Although more studies are still needed to determine whether the function of these brain wave patterns is the same across species, the results suggest that these REM and slow-wave, sleeplike patterns could date all the way back to the common ancestor of reptiles, birds, and mammals, Laurent says.The “provocative” findings also suggest that “there is something that goes on during sleep that is important to the function of all animals,” says Matt Wilson, a neuroscientist at the Massachusetts Institute of Technology in Cambridge. The lizards could be rehearsing the day’s events as they sleep, forming new memories of all the places they found a snack.
